The Role of miR-1 in the Progression of Gastrointestinal Stromal Tumors

The functional significance and molecular target of microRNAs in the progression of GISTs is investigated. MicroRNA expression array and Q-PCR detected differentially regulated microRNA-1 (miR-1) in gastrointestinal stromal tumors (GISTs) of different progression risk. The expression of Bcl-2, a repressing target by miR-1, in GISTs specimen was evaluated by western blot analysis and immunohistochemical staining. The result was correlated with that of TUNEL assay. Proliferation and cell death after oxygen-glucose deprivation (OGD)after in vitro transfection of miR-1 mimic or inhibitor was evaluated on HEK 293 cells. The very low risk (VLR) GISTs showed increased miR-1 expression and apoptosis, but decreased Bcl-2 expression. The high risk GISTs showed the reciprocal changes of miR-1, Bcl-2, and apoptosis as compared to those in VLR GISTs. Transfection of miR-1 mimic led to a decrease in Bcl-2 expression, less cell proliferation, and an increased of cell death after OGD in HEK 293 cells. Transfectants of miR-1 inhibitor showed increased Bcl-2 expression, more cell proliferation and a decrease of cell death after OGD. We conclude that miR-1 might work as a tumor suppressor by repressing the expression of Bcl-2 in GISTs of different progression risks. The epigenetic control by microRNAs plays an important role in GISTs progression.
