Identification and integrated analysis of differentially expressed CircRNAs reveal a potential Hsa_circ_0087378-miR-1260b-SFRP1 regulatory axis in ER-positive subtype breast cancer

Background: ER-positive subtype Breast cancer (BC) have low chemotherapy efficiency and may eventually result in disease progression and death. Recentlly, circRNAs are reported to be involved in multiple biological process in cancer. However, their roles in breast tumorigenesis are far from well investigated. This study focused on the systematic profiling and characterization of circRNA expression pattern of ER-positive subtype BC.
Methods: A human circRNA microarray was adopted to detect the differentially expressed circRNAs (DECs) in primary ER-positive subtype BC and adjuvant non-tumor tissues. Then, bioinformatics was utilized to depict the general characteristics of the DECs. Moreover, qRT-PCR was employed to validate the microarray analysis findings and predicted target genes in BC tissue samples and cells.
Results: In total, 3653 DECs were found in our paired samples. Moreover, most DECs were < 800 nt in length and chromosome 1 held the most DECs than any other ones. Go and KEGG pathway analysis showed most of the DECs were related to cancer-associated behaviors and the cell cycle, breast cancer and endocrine resistance pathways were key pathways involved. Furthermore, a circRNA-miRNA interaction network was constructed, and hsa_circ_0072304, hsa_circ_0087378 and hsa_circ_0087377 were validated to be down-regulated in ER-positive subtype BC. Additionally, the hsa_circ_0087378-miR-1260b-SFRP1 axis was proposed to be a key regulatory pathway in ER-positive subtype BC and a positive correlation was observed between hsa_circ_0087378 and SFRP1. Finally, survival analysis indicated that a worse overall survival in patients with a higher miR-1260b expression or a lower SFRP1 expression.
Conclusion: This study revealed the general expression characteristics of specific DECs in ER-positive subtype BC and hsa_circ_0087378 might be a promising candidate for further functional and mechanical investigation.
