Establishment of a novel method of evaluation drug efficiency in peritoneal micro-dissemination mouse model using luciferase assay

Peritoneal dissemination is a major form of recurrence in patients with malignant tumor in the abdominal cavity, and it is important as a factor with a poor prognosis. Animal experiments are essential for development new treatment, but previous animal models have several problems. We aimed at luciferase assay, and established a novel method in micro-peritoneal dissemination mouse model to evaluate tumorigenesis and drug efficiency.
BALB/c mice were intraperitoneally inoculated with colon26-luc, mouse colorectal carcinoma cell lines transfected pMSCV-luc. Luciferase assay was performed on day1, and mice observed luminescent spots were divided into no treat group, Cisplatin treated group, and Gemcitabine treated group. Drugs were injected intraperitoneally on day1 once. On day5 or day10, body surface and ex vivo luciferase assay was performed and mesenteric weight was measured to evaluate drug efficiency, and survival time of mice was inspected.
All mice expressing luminescence on day1 had peritoneal dissemination. As a result of luciferase assay on day5 or day10, there were significant differences with luminescence intensity by body surface luciferase assay and luminescent spots number by ex vivo luciferase assay between the groups treated or not. In each group, positive correlation was accepted between luminescence intensity and total spot number, and also mesenteric weight. Survival time of treated group was extended significantly.
We established a novel method to detect peritoneal micro-dissemination, and enabled to evaluate drug efficiency accurately in early time. Predicting survival time by luciferase assay, substantial shortening of experimental period could be expected. In future, developing of treatment against peritoneal dissemination, this novel method of evaluation drug efficiency and peritoneal micro-dissemination mouse model is useful for reduction of experimental animals and costs, and evaluation drug efficiency by short term.
